ABSTRACT Fresh corn is a highly appreciated vegetable in Brazil; this crop has high added value and profitability when compared to dry corn. Despite the consumer market demand for quality, cultivars are not launched every crop season and there is a small number of cultivars available to growers. This denotes a need for breeding programs focused on generating information and genotypes that can occupy this market niche. The objective of this work was to evaluate the progress of five cycles of intrapopulation recurrent selection in half-sib progenies of fresh corn, in the southwest region of Goiás. A randomized block experimental design was used, with evaluation of 64 progenies of corn half-sib progenies and selection intensity of 30% per cycle, with three replications, in the 2011-2012, 2013-2014, 2015-2016, 2017-2018, and 2020-2021 summer crop seasons. The half-sib progenies of fresh corn exhibited genetic variability through intrapopulation recurrent selection in the different selection cycles, which is still promising for further selections after five cycles, with evaluation of 64 progenies per cycle. The base population from crossings between hybrids for tropical and temperate climates showed to be promising for increasing the genetic base of fresh corn populations, with satisfactory gains over the five cycles, presenting mean heritability between the cycles of approximately 78.9% for relative chlorophyll index, 62.2% for plant height, 75.1% for ear insertion height, 70% for ear diameter, 56.6% for ear length, 71.4% for unhusked ear yield, and 71% for marketable ear yield.

RESUMO O milho fresco é uma hortaliça muito apreciada pelos brasileiros e, além disso, possui alto valor agregado e maior lucratividade, quando comparado aos grãos secos. O mercado consumidor apesar de exigente na qualidade, não é abastecido com novas cultivares a cada safra e com um número ínfimo sendo disponível aos produtores. Assim, existe a necessidade da criação de programas de melhoramento, com o intuito de gerar informações e genótipos capazes de suprir esse nicho de mercado. O objetivo do estudo foi avaliar o progresso da seleção em progênies de meiosirmãos de milho fresco, em cinco ciclos de seleção recorrente intrapopulacional, na região sudoeste de Goiás. O delineamento experimental utilizado foi o de blocos casualizados, com a avaliação de 64 progênies de meios-irmãos e intensidade de seleção de 30% por ciclo, com três repetições, em diferentes safras de verão (2011-2012, 2013-2014, 2015-2016, 2017-2018 e 2020-2021). As progênies de famílias de meios-irmãos de milho fresco apresentaram variabilidade genética por meio de seleção recorrente intrapopulacional nos diferentes ciclos de seleção, com avaliação de 64 progênies por ciclo. A população base de cruzamentos entre híbridos para climas tropicais e temperados mostrou-se promissora para aumentar a base genética de populações de milho fresco, com ganhos satisfatórios ao longo dos cinco ciclos, com herdabilidade média de aproximadamente 78,9% para o índice relativo de clorofila, 62,2% para altura de planta, 75% para inserção da espiga, 70% para diâmetro da espiga, 56,6% para comprimento da espiga, 71,4% para espigas empalhadas e 71% para espigas comerciais entre os ciclos.

Trichoderma harzianum isolates have been broadly used for biocontrol of plant diseases caused by fungi. Cladosporium herbarum is a common saprophyte and seed borne fungus, which is easy to manipulate under controlled conditions. It was chosen as a model to test the effectivity of seed treatments with T. harzianum. Common bean seeds (cv. Pérola) contaminated with C. herbarum were treated with conidial suspension (CS) and autoclaved filtrate (AF) of five isolates of T. harzianum and subsequently submitted to health and germination tests. The proportion of normal seedlings formed, the length of roots, hypocotyls and leaves, and total plantlet length, total plantlet biomass, root mass ratio (RMR), stem mass ratio (SMR), leaf mass ratio (LMR), aerial part/root system ratio (AP/RS) and leaf area were also evaluated. Isolates CEN289 and CEN290 (CS and AF) provided 66 to 77% of supression of C. herbarum on seeds and a higher number of normal seedlings as compared with control. It also yielded a higher total biomass of plantlets. Moreover treatment with isolates CEN289 and CEN290 increased root and stem length in the experiments with CS. Such results indicate the potential of T. harzianum for seed treatment and suggest that it should be further tested as control for seed borne fungal diseases and as a plant growth promoter. The better performance found for CEN289 and CEN290 confirms the variability in terms of biocontrol activity among strains of T. harzianum.

Biological control of seed-borne pathogens has shown to enhance germination and physiological quality of seeds. The objectives of this study were to evaluate the in vitro antagonistic effect of five Trichoderma harzianum isolates (CEN287, CEN288, CEN289, CEN290, and CEN316) against Fusarium oxysporum f. sp. phaseoli (Foxy) and test its potential use in seed treatment. Initially, pathogen and antagonists were grown in paired cultures at 25ºC, from which samples were assessed using scanning electron microscopy (SEM). Then, clean or Foxy-infected seeds were treated with conidial suspension of the antagonists. Percent of Foxy-infected seeds and normal seedlings were evaluated at seven and nine days of incubation, respectively. All but one Trichoderma isolate (CEN290) inhibited Foxy mycelial growth. SEM analysis revealed that only one Trichoderma isolate (CEN287) showed parasitic interaction with Foxy. Two isolates (CEN287 and CEN316) significantly reduced the Foxy incidence and enhanced seed germination, though less effective than the fungicide mixture (carboxin + thiram). A principal component analysis indicated the importance of volatile metabolites in reducing Foxy incidence on common bean seeds. CEN287 Trichoderma harzianum isolate formed a single group due to its increase in germination rate of Foxyinfected seeds.

A produção de conídios consiste em problema no estudo de Alternaria alternata do citros. Assim, este estudo objetivou comparar metodologias existentes para a produção de conídios de A. alternata por dois isolados de tangerina Ponkan, um de limão Cravo, dois de laranja Pêra e um de tangor Murcott. As metodologias empregadas foram a produção de conídios com 12 e 24 horas sob luz branca, avaliação com 24 e 48 horas após estressamento do micélio do fungo, choque térmico com imediato estressamento do micélio e avaliação com 24 horas, produção de conídios pelo emprego de tecido vegetal sadio e o emprego de luz negra ultravioleta. Produção satisfatória de conídios foi obtida com o isolado de A. alternata de tangor Murcott, a qual foi de 2,8 x 10(5) conídios mL-1, mediante emprego da técnica de estressamento da colônia e cultivo do fungo por 24 horas. Os empregos de luz branca (24 h) e negra ultravioleta promoveram expressiva produção de conídios por um isolado de tangerina Ponkan, a qual foi de 17,2 x 10(5) e 10,1 x 10(5) conidios mL-1 e por outro de tangor Murcott, a qual foi de 13,9 x 10(5) e 10,1 x 10(5) conídios mL-1, respectivamente. As outras metodologias analisadas neste estudo não foram capazes de induzir a produção de conídios em quantidade satisfatória. Com o emprego das técnicas de estressamento do micélio e a utilização de luz branca (24 h) e negra ultravioleta, tornou-se possível obter quantidades de conídios suficientes para serem utilizadas em testes in vitro (detecção de substâncias fungitóxicas)e in vivo (testes de patogenicidade), respectivamente.

Conidia production is a problem in the study of Alternaria alternata from citrus. Thus, this study aimed to compare existing methodologies for conidial production of A. alternata isolated from Ponkan tangerine (2 isolates), Cravo lemon (1 isolate), Pêra orange (2 isolates) and Murcott tangor (1 isolate). The methodologies used were conidia production with 12 and 24 hours under white fluorescent light, evaluation with 24 and 48 hours after applying fungal mycelium stress technique, cold stress followed by injury of mycelium and evaluation with 24 hours, using healthy vegetable tissue and the use of black fluorescent near ultraviolet (NUV) lamp. Satisfactory result was obtained with A. alternata isolate from Murcott tangor, with the production of 2.8 x 10(5) conidia mL-1, when fungal mycelium was stressed (Petri dish with 66.66% of fungi growth) and subsequently 24 h of growth. The use of white light (24 h) and black fluorescent NUV lamp also induced expressive conidia production by one isolate of Ponkan tangerine, which produced 17.2 x 10(5) and 10.1 x 10(5) conidia mL-1 and another of Murcott tangor, which produced 13.9 x 10(5) and 10.1 x 10(5) conidia mL-1, respectively. The remaining methodologies analyzed in this study were not able to induce conidia production in satisfactory quantity. The use of both mycelium stress technique and white light (24 h) and black fluorescent NUV lamp allowed the production of enough quantities of conidia to be used in vitro (detection of fungitoxic substances)and in vivo (pathogenicity test)assays, respectively.
